Dissecting method to collect worms from fish:
Using comprehensive dissection method Skrjabin, (1928): The fish was dissected starting from the cloaca to the mouth. Then separate the internal parts, each part will be separated and viewed directly with the naked eye or examined under a magnifying glass to find parasitic roundworms.
Enzyme hydrolysis method pepsin Jackson, (1981). For fish that detect roundworms, the fish fillet is further hydrolyzed with a mixture of water, pepsin and HCl 25%.
RFLP method for species identification:
PCR products were positive for Anisakis sp. used for species identification by restriction enzymes HinfI and HhaI.
Incubate the mixture at 37°C for 2 hours. Then electrophoresis on 2.5% agarose gel and stained with ethidium bromide. Read results under UV light. The results are compared with the species size table
